Abstract
In this study, to analyze the molecular mechanism of the formation of the lip-like petals and isolate the lip-like associated
genes, total RNAs were extracted from lip-like petals and normal petals in ‘Cang Shan Qi die’ by using plant total RNA extraction
reagent. Then mRNA were purified and transcribed into cDNA by SMART. After

I enzyme digested cDNA , using cDNA
fragments from the lip-like petals as tester and cDNA fragments from the normal petals as driver, Suppression Subtractive
Hybridization was applied to construct a differentially expressed cDNA library. Bioinformatics analysis shows there are 5 categories
of different differentially expressed genes in this cDNA library, which are MADS-box transcription factors, hypothetical protein,
non-matched protein in GenBank, storage proteins and others. 4 of them are B-class genes in MADS gene family. Phylogenetic
results show that 2 MADS genes belong to EDF genes, and the other 2 belong to PI genes. These MADS genes provide a basis for
cloning lip-like -associated genes in ‘Cang Shan Qi die’ as well as studying their function.
